I have lot of emails in my email queue I have tried deleting queue folder and even tried changing queue_old but again when I start KMS it again bring all those QUEUE and subfolders back how can I stop that some users sent an email to like 6K email address and now he wants me to stop that so I have no idea how to stop that, tried all the methods but queue SUBFOLDERS are keep coming back.

Please help.

Do you only allow authenticated users to your smtp server?
It smells like someone is abusing one of your accounts for sending spam.

barup is right, make sure you do NOT allow SMTP relay. I'd also look at the Mail log and see if you can determine where the emails are coming from. Could be a compromised email account used to send out spam emails. Could also be a client machine infected with a virus. Basically you'll have to determine from which account those emails are coming from, and lock down that account.
