Is KOFF supported for Remote Desktop users on Windows Server 2008 R2 or 2012 R2? I've seen some comments suggesting that it is, but I can't find anything definite.

Simple answer: yes.
Full answer: yes, but there are limitations.

KOFF is quite demanding on disk performance. So if more users are running KOFF on Remote Desktop with user data (i.e. KOFF data) located on the same physical disk, you can expect poor performance. It's not a big issue with small mailboxes (some 1GB or less), however once you have mailboxes several GB large, running more than 2 or 3 users with data located on same disk will be a problem. Not all the time, just when one of users makes some operation that requires lot of disk access (typically search), other users may experience Outlook not responding for tens of seconds.

I've heard about some issues with KOFF unable to run in Remote Desktop on Windows Server 2012 R2 at all, however it was probably configuration specific, we haven't be able to reproduce such issue.
