Can i delete or deactivate the SMTP-Standard-Rule?

You cannot disable basic functionality of the mailserver. Define rule(s) with higher priority.

OK.
First rule ist: "Use Relay, if sender is a local user."
Second rule should be: "Dont send if no local user.", but there is no Option for this.

The relay should only be used when a local user sends an email. Otherwise the SMTP should drop the mail. How do i do this?

in Webadmin select user -> Edit > Messages > the first checkbox.
If you want, this can be set for group of users:
Add or edit existing group of users -> Rights -> last checkbox
